虫草花多酚浸提工艺优化及抗氧化活性研究
Optimization of Extraction Technology of Total Polyphenols in Corhyceps Flowers and the Antioxidant Effects

摘要:
本文研究虫草花多酚浸提最优工艺,通过单因素试验和正交试验结合,以总多酚含量为优化指标。然后用石油醚、乙酸乙酯、正丁醇依次萃取,采用1,1-二苯基-2-三硝基苯肼自由基清除、总抗氧化能力检测试剂盒法、α-葡萄糖苷酶活性抑制三种体外抗氧化测定方法进行抗氧化活性测试。试验表明,影响多酚含量的提取因素为乙醇含量 > 提取温度 > 液料比 > 浸泡时间;虫草花中总多酚提取的最佳工艺为:液料比1:25 (g/mL),乙醇含量为80%,提取温度80℃,浸泡时间为1 h。虫草花的乙酸乙酯萃取物具有显著抗氧化活性,可以成为天然抗氧化活性化合物的良好来源。

Abstract: In this study, the extraction technology of total polyphenols from Corhyceps flowers was optimized through single factor experiment and orthogonal experiment. Then, the extraction was washed by petroleum ether, ethyl acetate and n-butanol. The effects of total polyphenols from different part in Corhyceps flowers on the scavenging of ABTS+ radicals, DPPH radicals and alpha-glucoside in-hibition were also studied. The result shows that the factors effected the Gallic acid content are the ethanol extraction > extraction temperature > solid-liquid ratio > extraction time and the best craft is using solid-liquid ratio 1:25, the content of ethanol is 80%, extraction temperature is 80˚C, the extraction time is 1 h. The ethyl acetate fraction of Corhyceps flowers, a strong ability to act as antioxidant, might be considered as a natural source of active compounds.
